- Press the plunger to the first stop, then immerse the tip into the liquid at a small angle—usually 20 degrees or less.
- Release the plunger slowly and smoothly to draw up the liquid. Wait about one second after the tip is pulled from the liquid.
- Dispense by pressing the plunger gently to the first stop, then continue to the second stop to expel the last drop.
- Hold the pipette vertically or slightly tilted; do not lay it flat with liquid in the tip.
Real talk: most pipetting errors come from releasing the plunger too fast, not pre-rinsing the tip, and touching the tip against the side of the tube in a way that changes the tiny volume. If you need the best accuracy, work with a calibration standard and practice the same rhythm every time.
Small labs often use the same pipette for years without checking the seal. A worn O-ring turns a 100 µL pipette into a guess. If your pipette doesn’t hold volume correctly, replace the seal or get it serviced. That’s cheaper than a failed assay.
